Methaqualone is classified as a sedative/hypnotic. It was originally synthesized in India to combat malaria but found to be ineffective. Methaqualone did prove effective as a sedative and was developed in the hopes of avoiding some of the adverse effects of the barbiturates, particularly their high capacity for addiction. Unfortunately, methaqualone was found to be just as addictive. Psysiologically, methaqualone is cumulative,and tolerance occurs rapidly in some individuals. In addition, it is extensively metabolized,at least 12 hydroxylated metabolites having been identified in the urine. The major metabolites are methaqualone-N-oxide, conjugate 4’-hydroxy-methaqualone,conjugated 2-hydroxymethaqualone.About 0.2% of methaqualone is excreted unchanged within 24 hours;40-50% of the methaqualone is excreted as metabolites within 72 hours,mostly as the glucuronide conjugates. The half-life for methaqualone averages 33 to 36 hours.It can be detected up to four days after administration.Side effects from chronic use of methaqualone are loss of motor coordination,walking into walls,ataxia,slurred speech,drowsiness and nystagmus.Severe acute overdose tends to produce muscle spasms, abnormally rapid reflexes,extreme muscle tension and restlessness.
